Problems solved by technology

Although a large number of genes related to stress resistance have been cloned from various plants, most of these genes can only increase a single resistance of plants, and cannot comprehensively improve the stress resistance of plants as a whole.

[0060] The seeds of Halimodendron halodendron were collected in the Tarim River Basin, Xinjiang. The 4-week-old bell thorn seedlings were soaked in 250mM NaCl for 3 hours and 6 hours, and the leaves were collected and treated with liquid nitrogen immediately, and stored in a -80°C ultra-low temperature refrigerator for later use.

[0061] Different stress treatments of materials: 250 mM NaCl, 20% PEG6000, 100 μM ABA solution, 100 μM GA3 solution, 100 μM BA solution, and 100 μM NAA solution were used to treat the bell thorn seedlings in hydroponics for 4 weeks. and placed in a 4°C incubator for low-temperature treatment, and samples were collected at 0, 1, 3, 6, 12, and 24 hours after treatment, and the processed materials were quickly frozen with liquid nitro...

Abstract

The invention discloses a halimodendron halodendron DREB transcription factor cDNA sequence, an expression carrier and applications thereof. In the invention, the DREB transcription factor cDNA sequence (SEQID NO:1) closely correlative to stress resistance is separated in a psammophyte halimodendron halodendron. The cDNA sequence is expressed in root, stem and leave of the halimodendron halodendron, and the expression is induced by aridity, high salt and low temperature environment and does not depend on the regulation of ABA. In the invention, an efficient plant expression carrier containingthe cDNA sequence is established and the plant expression carrier is translated into arabidopsis thaliana; the resistance analysis on aridity, high salt and low temperature environment is carried outon the transgenic arabidopsis thaliana; the relative physiological and biochemical indexes are measured; and the result indicates that the content of water, malondialdehyde, soluble sugar and pralineof the transgenic arabidopsis thaliana is obviously higher than that of the normal comparison sample, thus explaining that the cDNA sequence can improve the drought resistance and permeability stressresistance of the plant and is closely related to the stress resistance of the plant.

Description

technical field [0001] The present invention relates to a cDNA sequence related to plant stress resistance and the transcription factor encoded by it, in particular to the cDNA sequence encoding DREB transcription factor isolated and cloned from the psammophyte bell thorn (Halimodendron halodendron V.). The invention also relates to plant high-efficiency expression vectors containing the cDNA sequence and their application in improving plant stress resistance, belonging to the field of plant genetic engineering. Background technique [0002] Drought, salinity and low temperature are three major abiotic stress factors that affect the growth of terrestrial plants and limit crop yield. According to statistics, the world's arid and semi-arid areas involve more than 50 countries and regions, accounting for about 34.9% of the global land area. At present, among the 5.7 billion hectares of arable dryland used for agriculture in the world, about 70% of the arable land is degraded d...
