Embodiment 1
[0034] Example 1 Porcine Transmissible Gastroenteritis Virus (TGEV) Proliferation and Antigen Preparation
[0035]In this experiment, porcine testicular cells (ST) were selected as infected cells, and an appropriate amount of trypsin was added during virus infection to promote the adaptation and infection of the virus. The test results showed that the virus infection titer was significantly increased, and obvious cytopathic changes occurred. 1. Materials 1.1 TGEV isolates come from China Veterinary Drug Control Institute. 1.2 Newborn piglets 1-day-old colostrum-free piglets came from a pig farm in the suburbs of Beijing with no history of TGEV. 1.3 Porcine testicular cells (ST) were purchased from China Veterinary Drug Control Institute. 1.4 Cell culture medium MEM basal medium was purchased from GIBCO BRL Company, and neonatal calf serum was purchased from Hyclone Company. 2. Method 2.1 TGEV proliferates in pigs. After the 1-day-old piglets that did not eat colostrum were b...
Embodiment 3
[0044] Example 3 TGEV egg yolk antibody treatment test
[0045] 12 1-day-old piglets that did not eat colostrum were fed with milk, observed for 1 day, and randomly divided into 4 groups, namely: treatment group (4 pigs) for challenge, no treatment group (4 pigs), and no treatment group (4 pigs). Poison group (2 heads), no treatment and no attack group (2 heads). The virus used in the attack was ultrafiltered TGEV diluted 10 times with 0.01MPBS and then directly orally administered. The egg yolk antibody used in the treatment was a purified sample. The titer of the egg yolk antibody determined by the agar diffusion test was 1:64. and mortality rate to judge the therapeutic effect of TGEV yolk antibody.
[0046] The test results showed that the piglets in the two non-challenge groups grew normally without diarrhea symptoms, and whether the yolk antibody was fed or not had no effect on their growth; while the piglets in the two challenge groups all had severe diarrhea symptoms,...
