Abstract

The invention discloses a novel coronavirus vaccine candidate strain taking an influenza B virus as a vector as well as a construction method and application of the novel coronavirus vaccine candidate strain, and relates to the field of novel coronavirus vaccines. After an NS1 gene segment of an influenza B virus attenuated strain B / Yamagata / 16 / 88 is truncated and 110 amino acids are reserved, the NS1 gene segment is inserted into a receptor binding domain gene segment on a genetically synthesized novel coronavirus reference strain S protein, and the novel coronavirus vaccine candidate strain with the influenza B virus as the vector is obtained. The method comprises the following steps: constructing a recombinant plasmid NS110-RBD; a recombinant influenza virus rescued strain rIBV-NS110-RBD which takes an influenza B virus attenuated strain B / Yamagata / 16 / 88 as a skeleton and stably expresses a receptor binding domain (RBD) gene segment on a new coronavirus reference strain S protein is rescued by using a reverse genetics technology. The invention lays a foundation for the research on the packaging mechanism of the influenza B virus and the research and development of the novel coronavirus and novel influenza virus multi-combined vaccine.

Description

technical field [0001] The invention relates to the technical field of novel coronavirus vaccines, in particular to a novel coronavirus vaccine candidate strain with type B influenza virus as a carrier and a construction method and application thereof. Background technique [0002] The novel coronavirus has caused a severe outbreak of COVID-19 worldwide and was declared a pandemic in March 2020. According to the World Health Organization (WHO), as of October 2021, more than 200 countries around the world have reported 235,673,032 confirmed cases, including 4,814,651 deaths. [0003] The new coronavirus (SARS-COV-2) is the main pathogen that causes the new coronavirus pneumonia. It belongs to the Coronaviridae family and the βcoronavirus genus with the other two coronaviruses SARS-CoV and MERS-CoV that infect humans. The new coronavirus is an enveloped, positive, single-stranded RNA virus with a genome of 29903nts, composed of nucleocapsid protein (N), membrane protein (M), ...
