Embodiment 1

[0036] 1. Extraction of total RNA from Eriocheir sinensis

[0037] Extract healthy Chinese mitten crabs (about 40g each, purchased at the mouth of the Chinese mitten crab breeding pond in Baoying County, Yangzhou City, Jiangsu Province, collected by Huang Ying, collection date 2018.06.18) blood cells, heart, hepatopancreas, gills, muscles , gut, nerve and eye stalk tissue. In addition, 50 μL of Staphylococcus aureus and Vibrio parahaemolyticus (about 3×10 7Bacteria) were injected into the second step of the river crab, and the hepatopancreas tissues of different time periods (2, 6, 12, and 24 h) were extracted to analyze the changes of EsTNF expression after pathogenic bacteria stimulation.

[0038] (1) Add 1 mL of TRIzol reagent to 100 mg of tissue, fully homogenize in an ice bath; then transfer to a 1.5 mL centrifuge tube, and let stand at 15-30°C for 5 minutes;

[0039] (2) Add 200 μL of chloroform, shake vigorously for 30 seconds and let stand at 15-30°C for 5 minutes; ...

Abstract

The invention discloses a Chinese mitten crab tumor necrosis factor, the amino acid sequence of which is shown in SEQ ID NO.1. Also disclosed is the gene encoding tumor necrosis factor of Chinese mitten crab, the nucleotide sequence of which is shown in SEQ ID NO.2. Also disclosed is the application of Chinese mitten crab tumor necrosis factor in inhibiting Staphylococcus aureus or / and Vibrio parahaemolyticus. Also disclosed are recombinant vectors and recombinant engineering bacteria containing the coding gene of Chinese mitten crab tumor necrosis factor. The research results of the present invention show that the EsTNF protein can inhibit the growth of Staphylococcus aureus and Vibrio parahaemolyticus, and promote the clearance rate of bacteria in crabs. The invention discloses the application of the EsTNF gene in the immune defense function of the Chinese mitten crab, provides a theoretical basis for preventing diseases caused by pathogenic bacteria, and also provides theoretical guidance for healthy cultivation of river crabs.

Description

technical field [0001] The invention relates to Chinese mitten crab tumor necrosis factor, its encoding gene and application. Background technique [0002] Tumor necrosis factor (TNFs) is a class of powerful inflammatory cytokines involved in inflammatory response, apoptosis, cell proliferation and immune response. TNFs also function as effector molecules in host defense mechanisms by regulating cell death and survival. The TNF superfamily activates cell death and survival pathways, serving as a strong barrier against invading viruses. In mammals, the structures and functions of more than twenty TNF ligand superfamily member genes have been clearly defined. Most TNF ligand superfamily members are type Ⅱ transmembrane proteins with a conserved homotrimeric C-terminal TNF homology domain (TNF homology domain, THD). THD folds into an antiparallel β-sandwich structure and assembles into trimers with three receptor-binding sites per ligand forming grooves between adjacent subu...
