Abstract

The invention discloses a candidate marker vaccine strain with HQ06 epitope mutation for swine fever and an application of the candidate marker vaccine strain. According to the invention, a C strain is used as a skeleton, and three mutant viruses are constructed by single point mutation of three key amino acids 117F, 119G and 122P of E2 protein HQ06 antigen epitopes. The three mutant viruses constructed by the invention lose reactivity with a monoclonal antibody (mAb) HQ06 and retain reactivity with other antigen sites of the CSFV E2 protein, and biological characteristics of the three mutant viruses are consistent with those of a C strain; wherein the rHCLV-E2P122A can reach the growth titer of a parent virus in vitro, can induce an organism to generate an antibody aiming at the CSFV E2 and a neutralizing antibody aiming at the CSFV in rabbit and pig bodies, but loses a specific antibody aiming at the HQ06 epitope for inducing the organism to generate, and the rHCLV-E2P122A strain disclosed by the invention can be developed into a swine fever labeled vaccine.
