Embodiment 1

[0049] Example 1 Exploration of the Toxic Effect of TCDD on Glioblastoma

[0050] U87 cells were treated with 1×10 5 Cells / mL were inoculated into 96-well plates with 100 mL of cell suspension per well. Incubate overnight to allow cells to attach. Take out the 96-well plate, aspirate the original medium, and wash once with phosphate buffered saline (PBS). The experiment set up background control group, solvent control group and drug treatment group. Correspondingly, low-serum medium (1% serum concentration, no cells in the background control well), low-serum medium containing solvent dimethyl sulfoxide (DMSO) or different concentrations of TCDD was added to each well, and the volume of the solvent was maintained at 0.1%. Put the 96-well plate after adding the drug into the incubator to continue culturing for 24h or 48h. After reaching the end point of the culture, add 10 μL of CCK8 reagent to each well according to the instructions of the kit, put the well plate into the ...

Embodiment 2

[0052] Example 2. TCDD inhibits U87 cell migration

[0053] Cell migration ability was evaluated by scratch test. Use a marker pen to draw 3 straight lines horizontally on the bottom of the 6-well plate to locate the scratches. cells in 4 x 10 5 Inoculate 2 mL of cells / mL in a 6-well plate, culture overnight and wait for the cells to adhere to the wall. After the cells have completely covered the bottom of the well plate, scratch the cell monolayer with the tip of a 1 mL pipette to form a scratch. Choose the junction of the scratch and the marker line as the photo location. Wash with phosphate buffered saline to remove floating cells, then add low serum medium containing solvent DMSO or TCDD. Images were collected under an inverted microscope with a digital camera at 0-48 hours after scratching. A total of three independent repeated experiments were carried out, each concentration group in each independent experiment had 3 replicate wells, and each well contained 3 scratc...

Abstract

The invention relates to application of dioxin in inhibition of cell migration. Specifically, the invention relates to application of dioxin or a medicinal composition containing the dioxin to preparation of a medicine for inhibiting glioma cell migration. The invention finds for the first time that TCDD with specific concentration can inhibit migration of a glioblastoma cell line U87, and a new choice is provided for treatment of glioblastoma.

Description

technical field [0001] The present invention relates to the field of molecular biology, more specifically, the present invention relates to technologies related to cancer treatment. The invention mainly includes the application of dioxin in inhibiting cell migration, and particularly discloses the application of TCDD in inhibiting glioblastoma cell migration. Background technique [0002] Brain and central nervous system tumors are the third leading cause of cancer-related death in all cancer-related case studies. Among brain tumors, astrocytoma has a high incidence, accounting for 15.1% of primary central nervous system tumors and 46.1% of primary central nervous system malignant tumors. Among them, glioblastoma is the most common, which belongs to a highly aggressive primary glioma, and is also a grade IV tumor in the World Health Organization (WHO) tumor malignancy classification. [0003] The comprehensive treatment of glioblastoma includes surgery, radiotherapy, syste...
