Problems solved by technology

[0010] To sum up, in the prior art, although systems with integrated separation and distribution methods are moving towards the direction of one-step separation and distribution, they either adopt the method of splicing the separation and distribution system, or use a method similar to flow cytometry. Fluorescent labeling method, there are problems of cell transfer loss or cumbersome labeling, and there is still a long way to go from the ideal one-step, label-free single-cell preparation requirements

Abstract

The invention provides a one-step single cell separation and distribution system. The system comprises a liquid path plate, a surface acoustic wave chip, a first micro-fluidic chip and a second micro-fluidic chip, wherein driving gas is introduced into the liquid path plate; the first micro-fluidic chip and the surface acoustic wave chip are sequentially attached to the side surface of the liquid path plate; the second micro-fluidic chip is attached to the lower part of the liquid path plate; the surface acoustic wave chip is provided with three stages of sound fields which are used for promoting cells in a sample flow channel of the first micro-fluidic chip to be arranged and gradually screened; an image recognition area is arranged at the tail end of a flow channel of the first micro-fluidic chip and is used for carrying out unmarked image recognition on cells separated from the upstream; after the second micro-fluidic chip receives a single-cell signal of the image recognition area and triggers the single-cell signal, nano-liter liquid drops containing the cells are accurately sprayed out from a nozzle, and after a non-single-cell signal is received and triggered, the second micro-fluidic chip moves to the position above a waste liquid pool for performing spraying out. Cell acoustic separation, image recognition and single cell distribution are integrated, and one-step single cell preparation is realized.

Description

technical field [0001] The invention relates to the field of biotechnology, in particular to a one-step single-cell separation and distribution system. Background technique [0002] Circulating tumor cells (CTCs), a type of tumor cells that disseminate into the body fluid circulatory system in primary tumors or metastases, are responsible for the initiation of metastasis and the spread of cancer to distant places. As a "liquid biopsy specimen" representing the primary tumor, CTCs are used in the monitoring of metastasis and recurrence, prognosis judgment, curative effect evaluation and individualized treatment of patients with solid tumors. The analysis of CTCs is of great value. At the same time, cancer cells of metastatic and recurrent tumors have new somatic mutations compared with primary tumors, and in clinical practice, it is often difficult to obtain re-biopsies from metastatic or recurrent tumors, which leads to morbidity during treatment. The diagnosis is unclear. ...
