Abstract

The invention discloses an on-chip imaging microscopic system for observing behaviors of microorganisms. The microorganisms are injected into a micro-fluidic chip, and a capillary channel in the micro-fluidic chip can restrain the behaviors of the microorganisms, so that the microorganisms are ensured to stay in an observable range; an image sensor is adopted to carry out imaging on the microorganisms in the micro-fluidic chip, index-matching fluid in the micro-fluidic chip is blended to enable the refractive index of the index-matching fluid to be consistent with the refractive index of an outer layer material of the micro-fluidic chip, protective glass on the image sensor is disassembled, and the image sensor is tightly combined with the micro-fluidic chip, so that a problem of refractive index adaptation is effectively solved, and high-resolution imaging of the microorganisms in the micro-fluidic chip is realized; and in addition, a large visual field and high-resolution imaging canbe simultaneously achieved by the image sensor, so that the on-chip imaging microscopic system provided by the invention can be used for simultaneously restraining the microorganisms and observing the behaviors of the microorganisms with high flux.

Description

technical field [0001] The invention belongs to the technical field of biomedical microscopic imaging, and more specifically relates to an on-chip imaging microscopic system for observing microbial behavior. Background technique [0002] In the process of life science research, microorganisms such as nematodes have a short life cycle, small individuals, are easy to cultivate, and can be operated in large quantities. The number of cells that make up the body is relatively small, and exhaustive research on the morphology and lineage of the constituent cells can be carried out. Therefore, it is often used in research work such as morphological analysis and genetic analysis. However, due to the fast movement of microorganisms such as nematodes and their slender bodies, it is difficult to observe a complete nematode in the field of view when the magnification is adjusted enough to observe the internal details of the nematode when using a traditional optical microscope. This is n...
