[0044] Example 1 Correlation between the expression of microRNA in saliva samples and the risk of AMS

[0045] 1. Material and Specimen Collection Description

[0046] The saliva samples of AMS-susceptible persons were collected from AMS patients who entered the plateau rapidly before entering the plateau, a total of 75 cases. The saliva samples of normal people were collected from 49 cases of normal healthy people who entered the plateau rapidly before entering the plateau. The diagnosis of AMS was confirmed by the International Common Diagnostic Criteria-Lake Lewis Score. All the groups did not take any preventive medicine before taking saliva. A total of 10ml of saliva was collected from each sample in a sterile, enzyme-free 50ml centrifuge tube. The clinical case characteristics of the patients are shown in Table 1. This study has been approved by the Medical Ethics Committee of the Army Military Medical University (Third Military Medical University), and all specimens...

Abstract

The invention relates to a microRNA marker for screening saliva of acute high altitude reaction susceptible persons and application of the microRNA marker in preparation of a reagent or kit for screening the acute high altitude reaction susceptible persons. The invention further provides a kit for detecting hsa-miR-134-3p molecules and the hsa-miR-15b-5p molecules in the saliva of tested persons through a real-time fluorescent quantitative PCR method, and the onset risk of the acute high altitude reaction is predicted through the expression level of the kit. The microRNA in a saliva sample isused as a molecular marker, and the kit has the characteristics of high specificity, high sensitivity, noninvasiveness, convenience and the like, and is suitable for large-scale screening of the acutehigh altitude reaction susceptible persons in a plain.

Description

technical field [0001] The invention belongs to the field of molecular biotechnology and its application. The invention relates to a microRNA molecular marker for screening the saliva of persons susceptible to acute altitude sickness, and further relates to its application and kit. Screen the risk of acute altitude sickness by screening the expression levels of hsa-miR-134-3p and hsa-miR-15b-5p in saliva, so as to assess the susceptibility of the human body to acute altitude sickness and reduce the risk of acute altitude sickness. Background technique [0002] Acute mountain sickness (AMS) occurs in plain people who have lived in low-altitude areas for a long time, 1-3 days after entering the plateau quickly without adapting to the new environment, including headache, dizziness, fatigue, insomnia , Emotional restlessness, flatulence, diarrhea and vomiting and a series of symptoms, among which severe headache is a typical symptom of AMS. In geography, a vast area with an alt...
