Embodiment Construction

[0093] The present invention relates to methods of expanding a natural killer (NK) cell fraction for implantation into a subject while maintaining or enhancing cellular function in vitro and / or in vivo. In one embodiment, culturing NK cells in vitro with nicotinamide and / or other moieties of nicotinamide and NK cell growth factors promotes said production of NK cell populations for use as a therapeutic in vitro expanded NK cell preparation. The NK cell preparation comprises an expanded functional NK cell population having parameters suitable for infusion into a subject (e.g. NK cells and a reduced CD3+ T cell strongly amplified partially together). In particular in this regard, the present invention can be used to provide transplantable NK cell fractions and protocols for their use, which can be applied to cell transplantation and infusion for the treatment of cancer and other diseases. Non-limiting applications may include: allogeneic adoptive immunotherapy and combination i...

Abstract

Methods of expanding a natural killer (NK) cell fraction for transplantation into a subject are provided, and particularly, methods for providing transplantable NK cell fractions and protocols for their use, which can be employed for applications in cell transplants and infusions for treatment of cancer and other disease.

Description

technical field [0001] The present invention relates to the expansion of natural killer (NK) cells, the selection of the expanded NK cell population for transplantation into a subject in need and said suitable therapeutic use, and the in vitro expanded NK cell fraction for use in said clinical setting transplantation in the treatment of hematological malignancies. The invention also contemplates kits comprising said expanded NK cell fraction. Background technique [0002] Natural killer (also referred to herein as "NK") cells are lymphoid cells that participate in immune reactions. These cells have various functions, especially killing tumor cells, cells undergoing oncogenic transformation and other abnormal cells in a living body, and are an important part of the innate immune surveillance mechanism. Clinical experience with NK cell adoptive immunotherapy highlights the need for better approaches to simultaneously maintain effective and efficient expansion of NK cell popu...
