Abstract

The invention discloses a method for effectively improving the rooting rate of hybrid tree tissue culture seedlings. The method comprises the following steps: (1) enabling a rootless seedling which issubcultivated for at least seven generations to root; (2) cutting young stem tips of the rootless seedling after being sterilely filtered into a length of 1 to 2 cm, performing the dark treatment, and inducing; (3) performing primary rooting on the seedling induced in step (2); (4) after the primary rooting is completed in step (3), performing the dark treatment for 4 to 6 days, performing the secondary rooting, wherein a liquid-solid culture medium for the secondary rooting is prepared from 15 to 20 g / ml of perlite and 55 to 60 ml of liquid culture medium; and (5) performing the light illumination for 20 to 30 days after the secondary rooting. By adopting the method disclosed by the invention, the virus probability of the seed seedlings can be reduced; the cluster buds are induced at theproliferation phase, so that the proliferation times can be improved, and the proliferation times is greater than 6 to 8 times; the rooting rate of the tissue culture seedlings can be apparently improved, and the rooting rate is 97 percent; and the transplanting survival rate is 99 percent.
