Problems solved by technology

The analysis of genetic diversity and phylogeny of wild apple in Xinjiang is mainly based on SSR primers, but the target fragments amplified by SSR primers currently used for the analysis of genetic diversity in wild apple in Xinjiang are all non-specific amplification. The protein is not clear, and the specific sequence and biological role of the corresponding genetic differences between Xinjiang wild apple populations or individuals are difficult to determine. Using these SSR primers to analyze different populations and individuals of Xinjiang wild apples or cultivated apples, the fragments of genetic differences revealed It is difficult to determine what biological function and other issues

Abstract

The invention discloses specific SSR primers for malus sieversii and the cultivated apple and applications of the specific SSR primers. Based on the whole genome sequence of the cultivated apple, a functional gene sequence relevant to the stress resistance of apple is adopted as a template, and 15 pairs of SSR primers amplifying SSR target bands with specificity, stability and polymorphism in malus sieversii and the cultivated apple are screened, wherein the nucleotide sequences of the SSR primers are as shown in SEQ ID No.1-30. 8 pairs of SSR primers only capable of amplifying SSR target bands in the cultivated apple with specificity are also screened. The protein products coded by the target bands amplified by the SSR primers are known, and the SSR primers can be used for carrying out genetic diversity, polymorphis or phylogenetic analysis on the malus sieversii and cultivated apple, thus disclosing the genetic diversity and phylogenetic relationship of the malus sieversii and cultivated apple and the influences of the obtained genetic variation on the malus sieversii and cultivated apple in the aspects including the biological function.

Description

technical field [0001] The present invention relates to SSR primers, in particular to specific SSR primers designed based on the genome of cultivated apples that can amplify specific, stable and polymorphic SSR target bands in Xinjiang wild apples and cultivated apples. The present invention further relates to The application of the specific SSR primer in the analysis of Xinjiang wild apple and cultivated apple genetic diversity, polymorphism analysis or phylogenetic analysis belongs to the field of apple specific SSR primer and its application. Background technique [0002] Xinjiang wild apple (Malus sieversii (Ledeb.) Roem.) belongs to the Tertiary relict plant, which may be the ancestor species of modern cultivated apple, and is an important natural germplasm gene bank for the sustainable development of forest fruit industry. In recent years, due to the interference of human activities and the threat of pests such as Agrilus mali Matsumura, the natural population of wild ...
