The invention discloses a method for determining content of hesperidin in dried tangerine or orange peel by virtue of an ASE-HPLC (accelerated solvent extraction-high performance liquid chromatography) process and belongs to the field of chemical detection. The method is short in extraction time and high in detection accuracy. The method disclosed by the invention adopts an ASE process for extracting dried tangerine or orange peel powder, and collecting methanol extract liquor; and determining the content of hesperidin in the methanol extract liquor by adopting an HPLC process. The method disclosed by the invention can replace a pharmacopeia method for extracting the hesperidin in the dried tangerine or orange peel and determining the content of hesperidin.
