Problems solved by technology

Although the current research on anti-tumor peptides has achieved good results, almost all peptides are derived from nature or chemically synthesized libraries, and anti-tumor peptides isolated from humans are rarely reported. These peptides may have certain potential immune immunity. Abnormal reaction possible

[0023] 1. Peptide screening and modification

[0024] Preparation of serum samples: Collect about 5ml of fetal umbilical cord serum, let it stand at room temperature for 30 minutes, store at 4°C for 4 hours, then centrifuge (2000r / min, 10min), absorb the upper layer of serum and store it at -80°C until use.

[0025] Two-dimensional Chromatography: Size Exclusion Chromatography (SEC): Serum samples were diluted 10 times with PBS (pH7.4), and each injection was 100 μl. The mobile phase is 20mmol·L-1KH2PO4+100mmol·L-1NaCl, pH=7.0, flow rate 0.5ml·min-1. Detection was carried out at 280 nm with a UV detector. Segmentation is collected according to the chromatographic elution curve. Since the SEC separation mainly depends on the size of the separated molecules, the first segment of the serum is mainly the large molecular weight protein in the serum. This study mainly collects the components after 160min-180min and 200min-220min, and concentrates and lyophilizes the collected chro...

Abstract

The invention discloses an anti-tumor polypeptide and a preparation method and application thereof. The polypeptide is completely matched with the 7-19th amino acid residues of a segment of glutamyltranspeptidase (gamma-GGT) protein; the amino acid sequence is VLGLLAVVLVLVI and is named VI-13. In order to improve the solubleness, the sequence is reconstructed into VLGLRLAVRVLRVLRVI and is named VI-17. The polypeptide is obtained through a solid-phase synthesis method. An in vitro and in vivo pharmacodynamic experiment result shows that growth of human hepatocellular carcinoma cell line Hep3B can be significantly inhibited and the anti-tumor polypeptide has potential anti-tumor drug development value.

Description

technical field [0001] The invention belongs to the technical field of biomedicine, and specifically relates to the screening, synthesis and application of a polypeptide. The polypeptide of the invention has antitumor activity. Background technique [0002] Tumor is still an important factor threatening human health, and about 7 million people die from tumor-related diseases every year. Although surgery and chemotherapy can control the disease to a certain extent, they often cannot completely eliminate tumor cells, and a large number of patients still have recurrence and metastasis. Currently, chemotherapy is the most common means of blocking tumor progression. However, traditional chemotherapy has exposed a large number of disadvantages, such as poor specificity, prone to drug resistance, differences in drug distribution in vivo, and drug clearance rate. Therefore, the research and development of highly specific targeted drugs for tumors is quietly emerging. [0003] For...
