Embodiment 1

[0021]In the first ten days of September, select the fruit pods of Goodyera repens (L.) R.Br. that grow naturally in the wild and have no pests and diseases, pick them before the pods are ripe but not cracked, and bring them back to the laboratory, and wash the pods with gauze surface. Disinfect the surface of the preliminarily treated fruit pods with 75% alcohol for 20-30s, rinse them with sterile water for 3 times, and wash them with 0.1% HgCl 2 The aqueous solution is sterilized for 10-15 minutes, and washed three times with sterile water repeatedly under aseptic conditions, and the water on the surface of the fruit pods is blotted dry with filter paper. The pods are then cut open with a sterile scalpel and the seeds scattered into the medium (1). About 1 week after inoculation, the seeds began to germinate one after another, and about 7 weeks later, leaf primordia differentiated. Then inoculate the small leaf orchid seedlings with 2-3 leaves on the medium (2), bud differ...

Abstract

The invention provides an in vitro preservation method of near threatened plant Goodyera repens with considerable medicinal value. The method includes: selecting field well growing Goodyera repens fruit pods without pest and disease damages, conducting surface disinfection treatment, then breaking the fruit pods, inoculating the seeds in a seed germination medium to obtain sterile seedlings, and transferring the seedlings into a clumpy bud proliferation medium, conducting clumpy bud proliferation to obtain clustered shoots, carrying out rooting and in vitro preservation medium induction, and then transferring a seedling-carrying culture bottle into a culture condition at 13+ / -2DEG C for culture. Through the steps of field acquisition of explants, seed germination, clustered shoot induction, in vitro preservation, recovery growth of preserved seedlings and the like, the method provided by the invention establishes a rapid propagation and in vitro preservation system of Goodyera repens, and the subculture cycle is up to about 3 years. The in vitro preservation method of Goodyera repens provided by the invention provides important theoretical sources and technical support for development, utilization and protection of Goodyera schlechtendaliana plant germplasm resources.

Description

[0001] Technical field: [0002] The invention relates to the field of plant biotechnology, in particular to a method for in vitro preservation of the plant Goodyerarepens (L.) R.Br. . [0003] Background technique: [0004] Goodyerarepens (L.) R.Br.) is a herbaceous plant of the Orchidaceae Variegated Orchid. Its plant height is about 10-25 cm. White stripes, light green on the back, flowering period from July to August, like to be born in the forests of hillsides and valleys at an altitude of 700-3800 meters. Although the wild resources of this species are distributed in most parts of my country, this species has been included in the "Red List of Species in China" as a near-threatened level. Orchid japonicus has high medicinal value. The whole herb is used as medicine to invigorate the lungs and kidneys, relieve swelling and relieve pain, and is used for bronchitis, insect bites and joint pain. At the same time, the small variegated leaf orchid has exquisite leaves and exqu...
