[0023] Example 1 Isolation and identification of strains

[0024] Collect the activated sludge from the sewage treatment plant, weigh 5 g of the activated sludge sample into a 150 mL Erlenmeyer flask containing 50 mL of sterile water, and cultivate it at 30°C and 140 rpm for 3 days, then take 5 mL of the sludge suspension and add 100 mL PAEs (containing 50 mg / L DMP, DEP, DBP, DOP and DEHP respectively) in the above MSM medium. After culturing at 30°C and 140 rpm for 7 days, 1 mL inoculum was continuously enriched and transferred 10 times each time, and the content of PAEs in the medium was correspondingly increased to 800 mg / L. Then the 10 times acclimatized culture solution was diluted 10 3 ~10 5 Spread on LB solid plates and incubate upside down at 30°C for 1-3 days. After a single colony grows on the plate, pick a single colony and streak it several times for purification, and obtain a strain of bacteria, numbered J-1. Then the strains were inoculated on LB solid ...

Abstract

The invention belongs to the technical field of environmental pollutant biological treatment, and specifically discloses microbacterium sp. J-1 used for degrading a plurality of phthalic acid esters (PAEs). The microbacterium sp. J-1 is preserved at China Center for Type Culture Collection (CCTCC) on 22th, Jan., 2015, and preservation number is CCTCC No.M 2015055. The microbacterium sp. J-1 is capable of degrading a plurality of PAEs simultaneously; a problem that multi-effect bacteria in culture collection are insufficient is solved; the microbacterium sp. J-1 is widely distributed in the natural world; adaptability is high; and the invention also provides application research of the microbacterium sp. J-1 in polluted soil restoration, and technical support is provided for biological treatment of PAEs soil pollution.

Description

technical field [0001] The invention belongs to the technical field of biological treatment of environmental pollutants, and in particular relates to a new bacterial strain capable of efficiently degrading various phthalates at the same time, more specifically, a microbial strain capable of degrading various phthalates. Bacillus ( Microbacterium sp.) J-1. Background technique [0002] Numerous studies have shown that phthalic acid esters (PAEs) are highly toxic, teratogenic, carcinogenic, mutagenic, and reproductively toxic. PAEs are mainly used as plasticizers in the plastics industry, and have become one of the most common pollutants in the world. They are listed as priority pollutants by the US Environmental Protection Agency (EPA) and China Environmental Monitoring Station. According to the research results of the National Cancer Institute of the United States, dimethyl phthalate (DMP), diethyl phthalate (DEP), di-n-butyl formate (DBP), butyl benzyl phthalate ( Six ph...
