Embodiment Construction

[0021] The present invention is explained in detail below in conjunction with embodiment.

[0022] 1. Preparation of Sequences

[0023] According to the GeXP primer design requirements and the FMDV-Asia1 genome published by NCBI, select the conserved region to design specific primers, and form specific chimeric primers by adding universal primers, while the universal primer sequences belong to non-biologically derived nucleotide sequences , in addition to synthesizing the universal primer sequence, and adding a Cy5 fluorescent label to the 5' end of the upstream universal primer. The primer sequence of the present invention that can specifically amplify the foot-and-mouth disease Asia1 virus nucleic acid is: AGGTGACACTATAGAATAACTGCCTACCAGAAGCAACC, which is named FMDV-Asia1-F in the present invention; GTACGACTCACTATAGGGAAGTATGTCTCCGCACGCTTC, which is named FMDV-Asia1-R in the present invention. Universal primers used in the present invention: Cy5AGGTGACACTATAGAATA, named UWD-F...

Abstract

The invention discloses primers and kit for detecting an Asia1-type foot-and-mouth disease virus (FMDV) and a preparation method of the primers and the kit. The kit for detecting the Asia1-type FMDV comprises four amplification primers used for a GeXP multiplex gene analysis system. Related experiments show that by sequences of FMDV-Asia1-F and FMDV-Asia1-R, the nucleic acid of the Asia1-type FMDV can be specifically and rapidly amplified, specific bands are detected by virtue of DNA electrophoresis and the specificity is extremely high. The Asia1-type FMDV can be subjected to high-throughput specific detection by virtue of the GeXP multiplex gene analysis system which has sensitivity of 10<2> copies.

Description

technical field [0001] The invention relates to a primer, a detection kit and a preparation method that can be used for detecting foot-and-mouth disease Asia1 virus. Background technique [0002] Foot-and-mouth disease (FMD) is an acute and severe infectious disease of animals caused by foot-and-mouth disease virus (FMDV). Huge political and economic losses, so the World Health Organization (OIE) listed it as the first class A infectious disease. FMDV can be divided into seven serotypes, namely A, O, C, Asia1, SAT1, SAT2 and SAT3, each of which has many different subtypes, and there is no crossover between the serotypes There is only partial cross-immunity between subtypes of the same serotype. [0003] The typing and differential diagnosis of foot-and-mouth disease virus has always been a research hotspot. Because there is no cross-protection among the serotypes of FMD, there is only partial cross-immunity between the subtypes of the same serotype, which makes the diagno...
