Embodiment 1

[0045] Embodiment 1 Improving Rodwood "Edward" "(Abelia 'Edward Goucher') is the method for salt tolerance

[0046]1. Disinfection of materials: Take the young branches of Liudaomu "Edward", cut them into about 4-5cm stems with axillary buds, and first wash them in washing liquid with washing powder (washing liquid refers to the mass fraction containing about 0.5% dodecyl Sodium benzenesulfonate aqueous solution) for 5-6 minutes, then rinse with tap water for 2-3 hours, then cut off the unfolded leaves, and cut into about 1.5cm bud stem segments. On the ultra-clean workbench, soak in 75% alcohol solution for 20 seconds, then transfer to 0.1% mercuric chloride solution for sterilization for 8-10 minutes, and rinse with sterile water 5-8 times.

[0047] 2. Induction of adventitious buds: the sterilized stems were cultured on the medium of MS + kinetin 1.0 mg / L + indole butyric acid 0.1 mg / L + 3% sucrose + 0.1% NaCl 40-50 days. During the bud induction process, the stem segment...

Embodiment 2

[0057] Example 2 The method for improving the salt tolerance of Abelia grandiflora (scientific name: Abelia grandiflora "Francis Mason")

[0058] 1. Disinfection of materials: Take the young branches of Hexaphyllum mosaicus, cut them into about 4-5cm stems with axillary buds, and first wash them in washing liquid with washing powder (washing liquid refers to dodecylbenzene with a mass fraction of 0.5%) sodium sulfonate aqueous solution) for 5-6 minutes, then rinse with tap water for 3 hours, then cut off the unfolded leaves and cut them into 1.5cm budded stem segments. On the ultra-clean workbench, soak in 75% alcohol solution for 20 seconds, then transfer to 0.1% mercury solution for sterilization for 8-10 minutes, and rinse with sterile water 6-8 times.

[0059] 2. Induction of adventitious buds: the sterilized stems were cultured on the medium of MS + kinetin 1.0 mg / L + indole butyric acid 0.1 mg / L + 3% sucrose + 0.1% NaCl 40-50 days. During the bud induction process, the...

Abstract

The invention discloses a method for improving Abelia seedling salt tolerance by oriented induction. The method comprises the steps of: (1) material sterilization, (2) induction of adventitious buds, (3) bud differentiation and subculture, (4) rooting culture, (5) seedling hardening, (6) seedling transplanting, and (7) seedling management. The method provided by the invention makes use of tissue culture and container seedling raising to add NaCl with different concentrations into a differentiation proliferation medium, a rooting medium and a culture matrix for transplanted seedlings gradually, so that the seedling salt tolerance can be enhanced from about 0.2% to about 0.6% (normal growth can be realized when the NaCl content of an annual seedling matrix is 0.4-0.5%, and normal growth can be realized when the NaCl content of a biennial seedling matrix is 0.5-0.6%), and the fixed planting survival rate in saline-alkali soil can be over doubled.

Description

technical field [0001] The invention relates to the technical field of garden tree cultivation, and specifically refers to a method for obtaining hexagram seedlings and improving salt and alkali resistance by using modern seedling raising techniques. Background technique [0002] It is native to my country, and it is a deciduous shrub of the Lonicera family, and it is the most famous ornamental tree species with colorful leaves in my country. Rodaoki has strong adaptability, low temperature resistance, resistance to poor soil, and has a certain resistance to salt and alkali and drought. Liudaomu is one of the important landscaping tree species in the southeast coast of my country. However, the southeast coast is located in the subtropical zone and has a large annual rainfall. The salinized soil is gradually desalinated by rainwater and salt. The salt composition is mainly chloride, with a salt content of 0.1 %-0.8%. Contents of the invention [0003] Although Rodaria has ...
