The method comprises the following steps: (1) tributyl phosphate is used as an extracting agent for extracting the alpha-arbutin fermentation solution, and further an extract phase and a raffinate phase are separated, wherein the volume ratio of the extracting agent to the alpha-arbutin fermentation solution is 1 to (0.5-2) and the extract phase contains the hydroquinone; (2) stripping the extract phase obtained in the step (1) by utilizing a strip liquor, so as to separate the hydroquinone from the extract phase conveniently, wherein the strip liquor is a basic solution or an acid solution and the volume ratio of the extract phase and the stripping liquor is 1 to (0.01-0.5). With adoption of the method, the hydroquinone in the alpha-arbutin fermentation solution can be extracted efficiently at low cost, the impurity removing rate of the hydroquinone can be improved, the highly purified alpha-arbutin can be obtained, the emission of wastewater is reduced, and the method has an important significance on improving the economical efficiency of alpha-arbutin prepared through a biologic method.
