The method sequentially includes following steps: using rhizomes for asexual reproduction: digging out rhizomes of the wild atractylodes japonica, planting the rhizomes in an experimental field for asexual reproduction to enable the wild atractylodes japonica to adapt to ecological environment of domestic planting; and using seeds for sexual reproduction: sowing quality seeds obtained by directional strain selection for sexual reproduction in the next year; using rhizomes for asexual reproduction; and using the rhizomes of the previous year for cultivation for asexual reproduction in the third year; and allowing domestication characters to stabilize in the fourth year. By the method, the wild atractylodes japonica is domesticized into a novel variety suitable for garden planting and pastoral ecological environments, and high quality and high yield are realized.
