Problems solved by technology

In addition, the expression of exogenous Bt insecticidal protein in cotton plants showed a decreasing trend during the occurrence of the second, third and fourth generations of cotton bollworm. The overall decline caused the error in the evaluation

Embodiment 1

[0025] (1) During the occurrence period of the second, third and fourth generations of cotton bollworm, collect 10-20 leaves of the second fully expanded cotton top, respectively; use non-transgenic cotton varieties as control materials;

[0026] (2) Put the collected fresh leaves in the experimental box with fresh-keeping effect, one piece in each box;

[0027] (3) Connect one 1d-instar newly hatched larvae of cotton bollworm in the experimental box with leaves, tighten the lid of the box, and place it at room temperature;

[0028] (4) Investigate the number of dead larvae and age of cotton bollworm larvae on the 5th day after inoculation; if the tested insects die, the age is recorded as 0;

[0029] (5) Carry out statistical analysis, calculate the age inhibition rate of cotton bollworm, and determine the resistance level index based on the inhibition rate of the second, third and fourth generation cotton bollworm occurrence stages (see Table 1);

[0030] (6) The experiment...

Embodiment 2

[0037] In 2012, the existing cotton bollworm resistance identification method "Environmental Safety Testing of Transgenic Plants and Their Products

[0038] Insect-resistant cotton Part 1 (Announcement No. 953-12-2007 of the Ministry of Agriculture), the bioassay was carried out on the four cotton materials A2, B4, C4 and D2 in Table 2. The experimental procedures and identification results are as follows;

[0039] 1. Sampling method

[0040] During the second, third, and fourth generation peaks of the target pests, the third cotton leaves unfolded from the top were collected from the transgenic insect-resistant variety field and the insect-susceptible control variety field, and 20 cotton leaves were randomly collected from each plot per treatment.

[0041] 2. Operation

[0042] Add 20 mm of agar medium into a large test tube (35 mm×120 mm), insert the leaves (petiole reserved) into the medium to keep fresh, put one leaf in each test tube, and pick up 5 1-day-instar larvae of...

Embodiment 3

[0068] In 2012, the existing detection standard for exogenous protein expression in transgenic Bt cotton "Technical Specifications for Detection of Exogenous Protein Expression in Transgenic Plants and Their Product Components Detection of Insect-resistant Transgenic Bt Cotton (Ministry of Agriculture Announcement No. 1485-14-2010)" The exogenous protein expression levels of four cotton materials A2, B4, C4 and D2 were detected, and the detection results are as follows:

[0069] 1. Detection method

[0070] The test operation was strictly carried out according to the instructions of the detection kit, and the detection kit was the Cry1Ac / Cry1Ab detection kit produced by Envirologix Company of the United States.

[0071] 2. Test results

Abstract

The invention relates to the field of biological control, in particular to an evaluation method for cotton bollworm resistance of Bt (bacillus thuringiensis)-transgenic cotton. The method comprises the following steps: leaves are collected in emergence periods of a second generation, a third generation and a fourth generation of cotton bollworms; and non-transgenic cotton is adopted as a control material; the leaves are placed in a box; hatched larval cotton bollworms are inoculated; the dead quantity and instar of the larval cotton bollworms are investigated after the hatched larval cotton bollworms are inoculated; statistical analysis is conducted; and the instar inhibition rate of the cotton bollworms is calculated. According to the method, the sample quantity is relatively less; only one cotton bollworm can be inoculated, so that labor is saved and cotton bollworm tests are reduced, that is to say, the labor and the time are saved. According to the method, evaluations are conducted separately on the emergence periods of the second generation, the third generation and the fourth generation of the cotton bollworms, so that impacts and effects of foreign protein on the cotton bollworm resistance in the cotton growth and development processes can be reflected accurately, and the life of the cotton capable of resisting the cotton bollworms is prolonged. With the adoption of the method, the use quantity of the cotton bollworms is reduced greatly; and the method is easy to operate, simple, accurate, objective and reliable.

Description

technical field [0001] The invention relates to the field of biological control, in particular to a method for evaluating resistance to cotton bollworm of transgenic Bt cotton. Background technique [0002] In the 1990s, the cotton bollworm broke out and caused damage year after year, which brought a huge threat to my country's cotton production. The loss of cotton production reached 40% to 60%, and some cotton areas reached more than 80%, or even went out of production. The national cotton loss exceeded 40 billion yuan. In 1997, transgenic insect-resistant cotton was widely used in my country, which effectively controlled the outbreak of cotton bollworm and promoted the sustainable and stable development of cotton in my country. By 2006, the area of insect-resistant cotton in the country had reached 3.5 million hectares, accounting for 66% of my country's cotton area, of which the area of insect-resistant cotton in the Yellow River Basin accounted for more than 98%, and...
