Embodiment Construction

[0034] The present invention will be described in more detail below through specific embodiments and with reference to the accompanying drawings. It should be understood that the examples described below are only for illustration rather than limitation of the present invention.

[0035] One, the preparation of the cDNA of eucalyptus

[0036] Fresh eucalyptus (Eucalyptus urophylla x grandis cv DH32-29) leaves and petals were selected according to the method adopted by Guo at al. 2008, frozen in liquid nitrogen, and then stored in a -80°C refrigerator for later use.

[0037]Use cetyltrimethyl ammonium bromide (CTAB) method to extract tissue total RNA, specific implementation: 1) Preheat 15 ml CTAB extraction solution in a 65°C water bath; 2) Grind in liquid nitrogen for 2-3 1 gram of eucalyptus frozen tissue; 3) Transfer the sample to a centrifuge tube with CTAB extract, vortex vigorously at room temperature at 100-300 rpm for 30 seconds, and then bathe in 65°C water for 4-5 mi...

Abstract

The invention relates to sequences and functions of two eucalyptus genes (namely a WPGS3 gene and a WPGS4 gene) used for increasing plant biomass, belongs to the technical field of biology. At first, RNA (Ribonucleic Acid) in eucalyptus is extracted and is reversely transcribed to cDNA (complementary DNA), and then is subject to deep sequencing to obtain full-length cDNA sequence library of eucalyptus, the similarity comparison is carried out between the sequence of the cDNA sequence library of eucalyptus and amino acid sequences of arabidopsis AtGRF1 protein and arabidopsis AtEBP1 protein published by NCBI (National Center of Biotechnology Information) to obtain the full-length cDNA sequence of the WPGS3 gene and the full-length cDNA sequence of the WPGS4 gene, and finally, the full-length cDNA sequence of the WPGS3 gene and the full-length cDNA sequence of the WPGS4 gene are led into arabidopsis for overexpression. As a result, the biomass of transgenic plants is remarkably improved, and the functions of the two xylophyta genes in growth of dicotyledon are determined through a plant molecular biotechnology method at first.

Description

technical field [0001] The invention relates to the fields of plant genes, encoded proteins and applications, in particular to the WPGS3 gene and WPGS4 gene and their encoded proteins derived from eucalyptus and their application in increasing plant biomass. Background technique [0002] Eucalyptus is the general name of Eucalyptus in the family Myrtaceae. It is an excellent tree species that integrates ornamental, timber, pulp raw materials, and medicinal uses. Due to the long growth cycle of forest trees and high genetic heterogeneity, many traits are quantitative traits controlled by polygenes. Conventional breeding methods are difficult to meet the requirements of directional breeding of new eucalyptus varieties. Therefore, people place their research focus and success on genetic engineering technology. Solve the difficult problems of conventional eucalyptus breeding, and accelerate the process of breeding new varieties of high-quality and high-yielding eucalyptus. Fore...
