Problems solved by technology

Orthotopic transplantation of tumor tissue mass needs to prepare the tumor in advance, and it is time-consuming and laborious. It is impossible to study the mechanism of early tumor development because the primary tumor is not formed in situ

Embodiment 1

[0058] A kind of preparation of microencapsulated human pancreatic cancer tumor cell MiaPaCa-2, comprises the steps:

[0059] (1) Preparation of human pancreatic cancer tumor cell MiaPaCa-2 suspension

[0060] in CO 2 In the incubator, the human pancreatic cancer tumor cell MiaPaCa-2 was cultured in Dulbecco’s Modified Eagle Medium (DMEM) containing heat-inactivated fetal bovine serum, and the temperature was controlled at 37 °C, CO 2 The human pancreatic cancer tumor cell MiaPaCa-2 was grown and cultured under the condition of 5% concentration;

[0061] Take 20ml of human pancreatic cancer tumor cell MiaPaCa-2 in the logarithmic growth phase during the above culture process, discard the upper culture medium, add 4°C, pH7.2 PBS to wash three times, discard the supernatant, and then add 0.25% pancreatic Enzyme 4mL digestion, placed in 37 ℃, CO 2 The concentration is 5% CO 2 In the incubator, take it out after 5min, add 3mL of DMEM culture solution, pour the resulting soluti...

Abstract

The invention discloses microencapsulated human pancreatic carcinoma cells, and a preparation method and application thereof. The preparation method comprises the following steps of: adding suspension with human pancreatic carcinoma cells into Matrigel-containing sodium alginate solution; spraying into a calcium chloride solution in high voltage electrostatic environment to obtain microspheres; and performing ion exchange reaction on the microspheres and a sodium citrate solution, and thus obtaining the microencapsulated human pancreatic carcinoma cells. The obtained microencapsulated human pancreatic carcinoma cells have uniform size and the diameter of about 420mm. Cells in microcapsules are well proliferated, and grow in a three-dimensional way. The microencapsulated human pancreatic carcinoma cells are applied to subcutaneous tumor and in-situ tumor animal models, and have the advantages of high modeling success rate and quick tumor growth compared with the traditional tumor cell suspension injection method.

Description

technical field [0001] The invention belongs to the field of medical bioengineering, and relates to a microencapsulated human pancreatic cancer tumor cell and its preparation method and application. The obtained microcapsules have controllable volume and uniform size, active proliferation of tumor cells in the microcapsules, and a high rate of tumor formation in animal models. Background technique [0002] Pancreatic cancer is a malignant tumor with an overall 5-year survival rate of less than 5%. Less than 20% of patients with pancreatic cancer have limited lesions and can be surgically removed. Despite the rapid development of chemotherapy and radiotherapy, it has little effect on prolonging the survival time of patients. Therefore, there is an urgent need for animal models that can be used for the development of pancreatic cancer. [0003] At present, most animal models of human tumor cells can be divided into subcutaneous, orthotopic and heterotopic transplantation...
