Abstract

The invention provides use of a young scape as an explant for quick propagation and transgenosis of Chinese sacred lily, namely use of an un-budded young scape in a bulb of Chinese sacred lily as an explant material for in vitro quick propagation and transgenosis. A culture method comprises the following steps of: taking an un-budded flower ball grown by the scape, with the length of 1.5 centimeters; cutting the white part on the lower half part of the young scape into slices of 1 to 1.5 millimeters, and upwards placing the morphological upper end into an adventitious bud induction culture medium; and culturing the slices under light for 12 to 16 hours every day at the culture temperature of between 23 and 25 DEG C. The scape for in vitro quick propagation has the characteristics of high propagation coefficient and the like, and can be used for quick propagation of excellent seed balls. The culture method has the advantages of readily available materials, more material sources and easy establishment of a sterile system with sterilization; and the explant has high-frequency regeneration capacity, the differentiation rate of the adventitious bud reaches over 95 percent after 30 daysof culture, and the coefficient of differentiation is over 18, so the scape can replace the currently used bulb disc and be used as the explant for transgenosis and quick propagation and solve the problems of difficult sterilization and low transgenosis efficiency caused by using the bulb disc.
