Problems solved by technology

However, the above studies all focused on reducing the lactose content in milk, which solved the problem of lactose intolerance when people drink milk, but did not obtain functional galactooligosaccharide milk while producing low-lactose milk

Embodiment Construction

[0027] After taking out Escherichia coli (Escherichia coli) TTBGLYWT CGMCC No.3078, after the LB solid slant medium is activated, take a ring of bacteria from the slant seed and inoculate it into the Erlenmeyer flask containing LB seed medium for cultivation: shaker speed 200r / min, temperature 37°C, culture 12H. The seed culture solution was inoculated into the fermentation medium according to a certain inoculum amount for fermentation and cultivation: the rotation speed was 200r / min, the temperature was 37°C, and the cultivation was carried out for 24 hours. The fermentation liquid is centrifuged to obtain the bacterium, the wall is broken and centrifuged, and the supernatant is taken to prepare enzyme to obtain β-glucosidase TtβGLY.

[0028] During the preparation method of oligosaccharide-containing low-lactose milk, combined with the existing method for preparing low-lactose milk, the above-mentioned β-glucosidase TtβGLY is added to fresh milk for hydrolysis, and after hig...

Abstract

The invention relates to a strain, a method for preparing beta-glycosidase TtbetaGLY by the strain and a method for preparing low lactose milk containing oligosaccharide by the beta-glycosidase TtbetaGLY. The invention provides Escherichia coli TTBGLYWT CGMCCNo.3078. The method for preparing the low lactose milk comprises a conventional method for preparing the low lactose mil, i.e. the enzyme is added into raw and fresh milk to carry out hydrolysis; and after sterilized at superhigh temperature, the milk is filled to obtain a finished product. The enzyme is beta-glycosidase TtbetaGLY and is prepared by fermenting and extracting the genetic engineering strain Escherichia coli TTBGLYWT. By adopting the technical scheme, a great deal of galactooligosaccharide can be synthesized when reducing the lactose content in the milk, wherein the hydrolysis ratio of the lactose can reach 95.24 percent at maximum, and the galactooligosaccharide occupies 37.13 percent of sugar in the milk at maximum.

Description

technical field [0001] The present invention relates to a strain, a method for preparing β-glucosidase TtβGLY by using the strain, and a method for preparing oligosaccharide-containing low-lactose milk by using the β-glucosidase TtβGLY. Background technique [0002] Milk and dairy products contain high-quality protein, digestible and absorbable fat, rich minerals and multivitamins, and also contain a variety of immune substances, enzymes, hormones and other biologically active substances with physiological activity regulating functions. The average mass fraction of total milk solids is 13.0%, of which 4.0% is fat, 3.4% is protein, 4.8% is lactose, 0.8% is inorganic salt, and 0.11% is Ca. Milk and dairy products are indispensable ideal foods for human beings to improve their nutrition and enhance their physical fitness. [0003] The World Health Organization lists the per capita consumption of dairy products as one of the main indicators to measure the living standards of a ...
