The method comprises the steps of: the strain activation of Rhodospirillum rubrum ATCC11170; the seed culture of the strain; the fermentation culture of the strain in incandescent light and natural daylight in a lighting culture room; the collection of the hydrogen till the production of hydrogen is stopped. Two phases of lighting culture of the invention can obviously improve the photosynthetic hydrogen production rate. The method of the invention is easy to be operated, has low cost and wide application prospect.
