Embodiment
[0070] To exemplify the ability to use hydrogels to deliver drugs to the posterior segment, a trial was designed and a contact lens was used to deliver drugs to the retina. Eyeglasses from each New Zealand white rabbit were treated with VEGF, followed by one eyeglass with prednisolone, leaving the other eyeglass as a control. VEGF is known to cause edema of the retina and prednisolone is known to interfere with this mechanism. The contact lenses were high water ionomer lenses (SOFTLENS 66, Bausch and Lomb, Rocheser, NY) with a water content of approximately 66%. Each lens is about 13mm in diameter.
[0071] Lens Preparation-
[0072] The lenses were dehydrated according to standard manufacturing methods. Lenses were soaked in 1 mg / mL aqueous solution of VEGF (Sigma) for 12 hours at room temperature. Then, to load the lenses with drug, similar lenses were soaked in 1 mg / mL prednisolone in water for 12 hours at room temperature.
[0073] A lens containing VEGF was placed on...
