[0031] ① Cloning and sequencing of the full-length coding region of the 1Bx14 subunit gene. In order to obtain the full-length coding region of the 1Bx14 subunit gene from the genome of the high-quality wheat variety Xiaoyan 54, we designed a pair of PCR primers P3 and P4 (P3, 5'-ATCACCCACAACACCGAGCA -3' (SEQ ID NO: 6); P4, 5'-AGCTGCAGAGAGTTCTATCA-3' (SEQ ID NO: 7)). Using this pair of primers and genomic PCR reaction, we obtained amplified fragments of the full-length coding regions of six high molecular weight glutenin genes in Xiaoyan 54 (the fragment lengths range from 1.9 to 2.5 kb). After cloning the above fragments into the pGEM-T easy (Promega) vector, more than 200 positive clones were obtained. 50 positive clones were selected from them and the insert fragments contained in them were sequenced.
