Embodiment 1

[0034] 1. Method

[0035]1. Construction of CircEXOC6B (hsa_circ_0009043) overexpression plasmid vector

[0036] 1) Overexpression plasmid construction

[0037] The overexpression plasmid vector was constructed by Novopro Company through gene synthesis. Plasmid structure such as Figure 4 As shown, the expression sequence of back circular frame+circEXOC6B+front circular frame is shown in SEQ ID NO: 4, wherein the expression sequence of circEXOC6B is the 664-1053 bp of SEQ ID NO: 4.

[0038] 2) Cell transfection

[0039] Cell transfection was divided into two groups, namely circEXOC6B overexpression plasmid group and empty plasmid group (PLVX-PURO) or NC group. 48h after transfection, DU145 and PC-3 cells were collected.

[0040] 3) Design circEXOC6B primers and verify their specificity

[0041] Firstly, the total RNA of collected cells is extracted, and then reverse transcribed into cDNA. Design specific primers for cirEXOC6B. The sequence is as follows: Forward: CCTCA...

Abstract

The invention relates to an application of circular RNA-circEXOC6B (Ribonucleic Acid-CircEXOC6B) in inhibiting metastasis of prostatic cancer. Low expression of circEXOC6B in prostate cancer cells and prostate cancer tissues is found for the first time, stable transfection prostate cancer DU145 and PC-3 cell lines of the over-expressed circEXOC6B are further constructed, cell Transwell migration experiments and invasion experiments find that the over-expressed circEXOC6B can significantly inhibit migration and invasion of prostate cancer cells, in addition, a nude mouse pulmonary metastatic tumor model is constructed, and the model is used for screening prostate cancer cells and prostate cancer tissues. It is found that when the circEXOC6B overexpressed DU145 cells are injected into nude mouse bodies through caudal veins, in-vivo prostatic cancer metastasis can be remarkably inhibited. Therefore, circEXOC6B can be used as a new therapeutic target for prostatic cancer, and reference is provided for development of prostatic cancer drugs.

Description

technical field [0001] The invention relates to the technical field of biomedicine, in particular to the application of circular RNA-circEXOC6B in inhibiting prostate cancer metastasis. Background technique [0002] Prostate cancer (PCa) is one of the most common malignancies in men. In my country, with the improvement of socioeconomic level and medical and health conditions and the popularization of prostate specific antigen (PSA) screening, the incidence of prostate cancer is on the rise, and the age of first onset has declined; at the same time, prostate cancer The age-standardized death rate has been on the rise from 2000 to 2011. In 2020, the American Cancer Society predicts that there will be 191,930 newly diagnosed prostate cancer cases, accounting for 21% of male tumors, ranking first in the incidence of male tumors; 33,330 deaths, ranking second in male cancer mortality. The pathogenesis of prostate cancer is complex, and early localized prostate cancer can be cure...
