Embodiment 1

[0023] A method for preserving spermatozoa of Stren's sturgeon, comprising the steps of:

[0024] (1) Mix the Sturgeon's sperm with 8 times the amount of antifreeze, and then pack them into sterile containers;

[0025] (2) Pre-cool the mixed solution of Stryn's sturgeon sperm after subpackaging in step (1) to 5°C, store it at a constant temperature for 15 minutes, then cool the mixed solution to -20°C at a cooling rate of -1°C / min, Store at constant temperature for 10 minutes, then cool the mixture to -50°C at a cooling rate of -3°C / min, store at a constant temperature for 5 minutes, then cool the mixture to -100°C at a cooling rate of -5°C / min, and store at a constant temperature 3 minutes;

[0026] (3) Put the mixed solution obtained in step (2) into liquid nitrogen, and it is ready.

[0027] The antifreeze in the described step (1) includes 1.8 parts of sodium chloride, 1.5 parts of potassium chloride, 6.5 parts of sodium citrate, 1.2 parts of sodium hydrogen phosphate, 0...

Abstract

The invention provides a preservation method and recovery method of Acipenser schrencki sperms. The preservation method comprises the following steps: (1) uniformly mixing Acipenser schrencki sperms with 5-10 times of an antifreezing agent, and then subpackaging the mixture into sterile containers; (2) pre-cooling the mixed liquid of the sperms of the Acipenser schrencki subpackaged in the step (1) to 3-5 DEG C, conducting preserving at a constant temperature for 10-20 minutes, cooling the mixed liquid to -15 DEG C to -20 DEG C at a cooling speed of -1 DEG C / min, conducting preserving at a constant temperature for 5-10 minutes, cooling the mixed liquid to -40 DEG C to -50 DEG C at a cooling speed of -3 DEG C / min, conducting preserving at a constant temperature for 1-5 minutes, cooling the mixed solution to -80 DEG C to -100 DEG C at a cooling speed of -5 DEG C / min, and conducting preserving for 1-5 minutes at a constant temperature; and (3) putting the mixed solution obtained in the step (2) into liquid nitrogen to obtain a finished product. According to the preservation method for the sperms of the acipenser schrencki, the sperms are subjected to cryopreservation in a subpackaging and cooling constant-temperature mode, so that the generation of crystals of the sperms in a crystallization dangerous zone of 0-40 DEG C is reduced, and the structural integrity of cells is ensured.

Description

technical field [0001] The invention belongs to the field of sturgeon breeding, and in particular relates to a preservation method and a recovery method of sturgeon sperm. Background technique [0002] Sturgeon's sturgeon belongs to Osteophytes, Atenopterygia, Sclerophyta, and Sturgeoniformes. It is mainly distributed in the Amur River (Heilongjiang, China) basin. Because of its advantages such as fast growth, strong disease resistance, and high economic value, it has been It has become a popular cultured species in China, accounting for 50% of China's total sturgeon production, and there is still an increasing trend. However, due to extensive breeding techniques and high breeding density, the frequency of disease outbreaks has increased, which has greatly hindered the healthy and sustainable development of sturgeon aquaculture. In recent years, scientific researchers have done a lot of work in the protection of Acipenser schneidi resources, artificial breeding, artificial ...
