Embodiment Construction

[0015] The sense strand and antisense strand of the siRNA capable of knocking down SRC-1 gene expression of the present invention are as follows:

[0016] Justice strand: CAGCAGUCAUAGUAAUUCUTT;

[0017] Antisense strand: AGAAUUACUAUGACUGCUGTT.

[0018] 1. The expression of SRC-1 is positively correlated with the malignant grade of clinical glioma

[0019] The expression of SRC-1 in brain glioma tissue chips (59 cases in total, 56 cases of glioma, and 3 cases of normal brain tissue) was detected by immunohistochemical method. The immunohistochemical results were as follows: figure 1 As shown in A. The expression of SRC-1 in normal brain tissue is very low, and the expression of SRC-1 is high in different degrees in glioma tissue, and the expression of SRC-1 is higher with the malignant grade of glioma. The immunohistochemical results of SRC-1 expression in 56 cases of gliomas with different malignant grades were counted. The results are as follows: figure 1 As shown in the ...

Abstract

The invention discloses a siRNA capable of knocking down expression of a SRC-1 gene. A positive-sense strand and an antisense strand are separately as follows: the positive-sense strand: CAGCAGUCAUAGUAAUUCUTT and the antisense strand: AGAAUUACUAUGACUGCUGTT. By knocking down the SRC-1, cell proliferation of spongioblastoma and tumor growth can be inhibited, and sensibility of spongioblastoma cells to temozolomide can be increased. The siRNA can be used as a novel drug for treating spongioblastoma.

Description

technical field [0001] The invention belongs to the field of biomedicine, and in particular relates to an siRNA capable of knocking down the expression of SRC-1 gene and its application. Background technique [0002] Glioma is the most common malignant tumor of the central nervous system, accounting for about 80% of primary malignant brain tumors. According to the statistics of the European Neuro-Oncology Association (EANO), the annual incidence of glioma in the world is about 6 / 100,000 . Glioblastoma (GBM) is the most malignant brain glioma (WHO, grade IV, 2007), accounting for 16% of primary intracranial tumors and 50% of brain gliomas. The 5-year survival rate is less than 5%. The important biological characteristics of GBM are aggressive growth, easy recurrence and intracellular heterogeneity of tumor tissue. At present, a comprehensive treatment strategy including surgical resection, standard-dose radiotherapy (60Gy) and concurrent chemotherapy with temozolomide (TMZ...
