Abstract

The invention discloses a device and testing method for imaging the transparent droplet in light sheet fluorescence microscopy. The imaging device comprises a light source reshaping module, a light sheet generating module, a sample control module and an image acquisition module. The light source reshaping module converts the circular light spot into elliptical light spot; the light sheet generating module produces a sheet beam from the elliptical spot; the sample control module controls the sample to move in direction vertical to the optical axis when the sheet beam illuminates on the sample,and the image acquisition module acquires the fluorescence signal excited from the moving sample at different positions to obtain the sequential three-dimensional images of the sample. The invented device can generate an elliptical light spot in short optical path to form a bright and thick light sheet, which makes the beam shape more suitable for enclosed in-situ imaging of liquid droplets in deep location. Meanwhile, since no silt is used for blocking the laser the utilization of laser energy increases by four times. The invention enables the enlargement of light aperture under wide view field and the reduction of the length of collecting end, with small size and high integration degree.
