The method includes a step of extraction, a step of milkvetch root-glossy privet fruit polysaccharide purification, and a step of performing sulfation modification through a chlorosulfonic acid-pyridine process to obtain sulfated milkvetch root-glossy privet fruit polysaccharides. By adopting product immunological competence as an index, L<9>(3<4>) orthogonal experiments are performed to optimize a reagent ratio, reaction temperature and reaction time which are therefactors, and determine optimum modification conditions in which the mass ratio of chlorosulfonic acid to pyridine is 1:8, the reaction temperature is 60 DEG C and the reaction time is 1 h, and the immunological competence of the sulfated milkvetch root-glossy privet fruit polysaccharides obtained under the optimum modification conditions is highest.
