The method comprises the following steps that the perimeter of an inner opening of each breeding box body is accurately measured, and an adhesive tape with the width of 6 cm or more is adhered at the position 2 cm away from the lower portion of a box opening and surrounds an inner opening of the breeding box body from any starting point for one circle; the surface of the adhesive tape is coated with toothpaste, wherein the thickness of the toothpaste is not smaller than 2 mm; a spacing of at least 5 mm or more is reserved between the toothpaste and each of the two sides of the adhesive tape; the toothpaste width is at least 5 cm. Escaping of leeches in the box body along a pond wall is completely eradicated, the opening of the box body is not covered by an escape-preventing net, and breeding operation is convenient.
