Embodiment Construction

[0019] This embodiment is a method for screening functional candidate genes that affect meat quality traits, specifically comprising the following steps:

[0020] 1 Materials and methods

[0021] 1.1 Collection of longissimus dorsi muscle samples from Wannan Hua pigs and Yorkshire pigs

[0022] The samples selected in this experiment were Wannan Huahua pig and Yorkshire pig. Two groups of pigs, 3 individuals in each group, were randomly selected, with a weight range of 100-131kg, a total of 6 individuals. Take the longest muscle sample from the back of the pig body (about 1kg, located at the last rib to the penultimate rib), put it in a cryopreservation tube, and put it into a liquid nitrogen tank for storage immediately. After collection, take it out and store it in a -80°C refrigerator for later use.

[0023] 1.2 Extraction of longissimus dorsi muscle DNA and construction of sequencing library

[0024] In this experiment, phenol: chloroform: isoamyl alcohol 25:24:1 / Agenc...

Abstract

The invention discloses function candidate gene for affecting the meat quality trait of a pig and a screening method of t function candidate gene. The function candidate geneisRXRG, GPI, CPT2 and / or GABARAPL1. The screening method comprises the following steps: (1), selecting longissimus muscles of two extreme pig breeds with large meat quality trait difference, extracting DNA, and independently creating databases; (2), performing whole genome DNA methylation sequencing on sample DNA databases by adoptingan MeDIP-Seq technology; (3), obtaining maps of whole genome DNA methylation levels of the two extreme pig breeds, and screeningdifferentialmethylationexpressed genes; (4), performing GO functional annotation and KEGG Pathway analysis on the differentialmethylationgenes, and screening out functional genes and regulatory pathways related to meat quality traits, such as fatty acid formation and decomposition; (5), performing DNA methylation and expression profileconjoint analysis through MeDIP-seq and RNA-seqtest results, and obtaining the function candidate gene of the meat quality trait of the pig. According to the method, the function candidate geneparticipatinginregulationof the meat quality trait of the pig is screened out, and a foundation is laid for the future deep research.

Description

technical field [0001] The invention relates to a functional candidate gene affecting meat quality traits and a screening method thereof. Background technique [0002] Pork quality traits are important indicators for animal economic protein production. Although a large number of studies on the genetic mechanism of pork quality traits have been carried out, the research on candidate genes and genetic mechanisms at the genome-wide level is relatively limited. The rapid development of DNA sequencing technology in the past 20 years has become an important means of studying genetic mechanisms. A large number of studies have shown that transcription factors, growth factor receptors and signaling pathway mechanisms are involved in the regulation process of meat quality traits. [0003] DNA methylation is one of the first epigenetic modifications of genes discovered, and previous reports have shown that methylated CpG dinucleotides repress gene expression by altering chromatin str...
