The manual picking and automatic replenishment system comprises multiple layers of goods shelves, guide rails, shuttle vehicles, goods shelf conveying lines, station conveying lines and a control device, wherein each layer of goods shelf is equipped with an alley which partitions each layer of goods shelf into multiple rows and multiple columns; the guide rails are arranged in the alleys; the shuttle vehicles move along the guide rails to convey goods to the goods shelves; each goods shelf conveying line is located at the deliver-in end of the alley on each layer of goods shelf and is used for conveying the goods to each shuttle vehicle; the station conveying lines are located beside the goods shelves and are used for conveying finished products; the station conveying lines are equipped with multiple manual operation stations; and the shuttle vehicles, the goods shelf conveying lines and the station conveying lines are all connected with the control device in a communication manner.
