Problems solved by technology

Compared with other noble metal nanoparticles, silver has the best plasmon enhancement effect in the visible light band, and gold has excellent biocompatibility and stability, and can be modified on the outer layer of silver nanoparticles, so Ag@Au core-shell nanoparticle Particles may generate strong and specific plasmonic circular dichroism responses, but the plasmonic chiral activity of Ag@Au core-shell nanoparticles is less studied

[0045] 1) Preparation of Ag@Au core-shell nanoparticles

[0046] Silver nanoparticles with a particle size of 12±3nm were synthesized according to the previously reported method, 1 mL was centrifuged at 16200 g for 10 minutes, concentrated 10 times, and redissolved in 100 μL ultrapure water. 100 μL of the silver nanoparticle solution was added to 1 mL of a 1% polyvinylpyrrolidone solution under uniform stirring, and then 100 μL of a 100 mM ascorbic acid solution was added thereto. The pH of the solution was then adjusted to 9.5. A 0.1 mM chloroauric acid solution was added to the mixed solution at a rate of 100 μL / min using a constant flow pump. After stirring for 1 hour the final product solution was centrifuged and redissolved in 100 μL of ultrapure water.

Abstract

A novel circulating tumor cell detecting method based on a chiral nanometer assembly is provided and relates to the technical field of biological treatment. The method is characterized in that a circulating tumor cell is over-expressed in HER2 than normal cells, and a chiral Ag@Au core-shell nanometer particle assembly modified with an HER2 aptamer is utilized to achieve specific recognition of the circulating tumor cell. The method includes steps of preparing Ag@Au core-shell nanometer particles, preparing the chiral Ag@Au core-shell nanometer particle assembly modified with the HER2 aptamer, specifically recognizing and detecting HER2 positive breast cancer SK-BR-3 cells by utilizing a fact that dissociation occurs when the chiral Ag@Au core-shell nanometer particle assembly is brought into contact with HER2, and specifically recognizing and detecting the circulating tumor cell in a biological sample by utilizing the chiral Ag@Au core-shell nanometer particle assembly .

Description

technical field [0001] The invention establishes a new method for detecting circulating tumor cells based on a chiral nano assembly, which belongs to the field of nano biosensing. Background technique [0002] Chiral nanometamaterials originate from the plasmon-exciton interactions between chiral molecules and metal nanostructures and the spatial arrangement of assemblies. Studying the dynamic plasmonic circular dichroism response of chiral nanomaterials is the key to obtaining new chiral metamaterials, and has potential research value in asymmetric catalysis, enantioselective separation and plasmonic device applications. [0003] Controlling the interaction between chiral metamaterials and light is the key to regulating chiral signals, and the localized electromagnetic field enhancement generated by localized surface plasmon resonance (LSPR) of plasmonic nanoparticles provides an effective research avenue for amplifying chiral signals. Compared with other noble metal nanop...
