The method comprises the following steps of measuring lengths of left peduncle 3, left peduncle 4, left tentacula and mantle of the sepiella maindroni requiring gender discriminating; calculating by the following discriminants, wherein for male, F1=223.42X3+165.85X4+7.951X5-108.06; for female, F2=151.48X3+92.71X4+19.19X5-60.03; in the discriminants, replacing X3 with the length of left peduncle 3 / mantle, replacing X4 with the length of left peduncle 4 / mantle, and replacing X5 with the length of left tentacula / mantle.
