Problems solved by technology

Among them, Freund's adjuvant is only suitable for laboratory use due to its high price, and is not suitable for large-scale clinical promotion, and it is too viscous and difficult to inject
The biggest disadvantage of white oil adjuvant is that it needs to be heated and refined before use, the process is complicated, and the oil phase and water phase are easy to separate (especially in a low temperature environment of 4 degrees Celsius), which is not conducive to long-term storag

Abstract

The invention relates to the field of veterinary biological products and is aimed at providing a veterinary vaccine immunologic adjuvant as well as a preparation and application method thereof. The adjuvant is prepared by mixing water-soluble calcium carbonate, polysorbate 80, 2-phenoxy ethanol, chitosan oligosaccharide, xanthan gum and deionized water, wherein the weight ratio of the water-soluble calcium carbonate to deionized water is 0.001-0.01%; the volume ratio of the polysorbate 80 to deionized water is 1-10%; the final concentration of the 2-phenoxy ethanol is 0.5-2.5% (volume ratio); the final concentration of the chitosan oligosaccharide is 0.5-2% (weight ratio); the final concentration of the xanthan gum is 0.1-2% (weight ratio). The adjuvant provided by the invention can enhance the identification ability of lymphocytes for the A, B and H antigens, promote the combination of multiple antigens with an antibody and enhance the immune response level and antigen presentation ability. The adjuvant is kept in a steady state at low temperature and room temperature; due to the antibacterial ability, secondary infection after immunization is avoided. The adjuvant is easy to prepare and thus can be stored for long time at room temperature. The cost of raw materials is low, and the adjuvant is suitable for large-area promotion and application.

Description

technical field [0001] The invention belongs to the field of veterinary biological products, and in particular relates to a method based on calcium carbonate, xanthomonas polysaccharide (trade name xanthan gum), chitooligosaccharide, 2-phenoxyethanol and polysorbate 80 (trade name Tween 80)-based veterinary vaccine immune adjuvant and its preparation and application method. Background technique [0002] An immune adjuvant is a non-specific immune enhancer, which can enhance the body's immune response to antigens or change the type of immune response when immunized with antigenic substances in animals or humans. Therefore, immune adjuvants are widely used in human and veterinary vaccines to prolong the retention time of antigens in the body, make the antigens fully contact with lymphatic system cells, stimulate the production of antibodies, and enhance the immune effect. [0003] The immune adjuvants currently used in veterinary vaccines include: Freund's adjuvant, white oil...
