The preparation method comprises the following steps: 1, successively carrying out extraction with water, condensation of extracted liquid, alcohol precipitation, centrifugation for removal of a precipitate and reduced pressure condensation for removal of ethanol so as to obtain extracted liquid, or successively carrying out heating extraction with 40 to 95% ethanol in a water-bath with a temperature of 80 DEG C, cooling, centrifugation and condensation for removal of ethanol so as to obtain the extracted liquid; and 2, extracting the extracted liquid with ethyl acetate, subjecting ethyl acetate extracted liquid to separation with a silica gel column, wherein eluents are dichloromethane-methanol with different volume ratios, collecting eluate obtained by using dichloromethane-methanol with a volume ratio of 50: 1 and recovering the organic solvent in the eluate so as to obtain the extract of the antineoplastic active components in the vietnamese sophora root.
