Embodiment Construction

[0022] The present invention will be further described below by taking epididymis fibroblasts and kidney fibroblasts of newborn Guangxi Bamaxiang pigs as examples. It should be noted that this embodiment is only a specific embodiment of the present invention. Obviously, the present invention is not limited to the examples. All deformations that can be directly derived or associated by those skilled in the art from the content disclosed in the present invention should be considered as the protection scope of the present invention.

[0023] 1. Fibroblast isolation and culture

[0024] Guangxi Bamaxiang pigs within 1 to 2 weeks of age were obtained, epididymis and kidney tissues were aseptically collected, and Guangxi Bamaxiang pig epididymis fibroblasts and kidney fibroblasts were isolated by primary culture according to the above-mentioned procedures.

Abstract

The invention discloses a method for non-freezing long-term cell preservation. The method comprises the following operation steps: (1) primary culture, (2) secondary culture, and (3) non-freezing long-term cell preservation, namely contact inhibition. The invention has the beneficial effects that the method has the advantages of lower cost, less pollution and high operation convenience and reliability compared with the conventional cell preservation based on a liquid nitrogen container. Particularly for some special projects (for instance, rare and precious genetically modified cells), after the rare and precious genetically modified cells are subjected to a batch of somatic cell cloning, the implementation of large-batch embryo production, namely embryo transplanting, needs to be determined by further verifying the in-vivo development condition of an embryo. If the special cells during pregnancy duration are subjected to freezing preservation, the survival rate can not be guaranteed, and meanwhile, contamination probabilities are increased; and if the conventional culture transfer is subsequently carried out, the probability of cytometaplasia and cell contamination can be increased to further influence the in-vitro normal development of the embryo. Thus, the method for non-freezing long-term cell preservation is an alternative method.

Description

Technical field: [0001] The invention belongs to the field of life sciences, in particular to a method for non-freezing long-term preservation of cells. Background technique: [0002] So far, no report identical or similar to the present invention has been found. Invention content: [0003] The purpose of the present invention is to provide a non-freezing method for long-term preservation of cells. [0004] The technical solution of the present invention to solve the above-mentioned technical problems is as follows. [0005] The operation steps of this method are as follows: [0006] A method for non-freezing long-term preservation of cells, the operation steps of which are as follows: [0007] 1) Primary culture [0008] Primary culture and separation of fibroblasts using the micro-liquid tissue block adherence method, aseptically collect animal living tissue blocks, quickly rinse with 75% alcohol for about 60 seconds, and immediately put them in DPBS containing 3 tim...
