Embodiment Construction

[0029] The present invention is described in further detail below, which is an explanation of the present invention rather than a limitation.

[0030] 1. Microbiological sample collection

[0031] The new actinomycete species CCTCC M 209033 of the present invention is isolated from soil polluted by heavy metals, and the soil polluted by heavy metals is collected from the lead-zinc mining area of Liulin Town, Huixian County, Gansu Province (106°07′44.3″E, 33°54′ 10.3″N, 1049 meters above sea level).

[0032] Collect about 500g of topsoil soil samples, divide them into sealed plastic bags, and air-dry them at room temperature. Use a No. 18 sieve (aperture 1mm) to remove larger gravel and animal and plant residues, collect soil samples, and store them in a dry place at 4°C. spare.

Abstract

The invention discloses a novel species of actinomyces resisting heavy metal lead and application thereof. The bacterium species belongs to the streptomyces, is named as streptomyces plumbiresistens sp.nov, and is applied to the biological repair of an environment with heavy metal lead pollution. System research is carried out to a bacterial strain CCTCC M 209033 with higher lead-resisting characteristic by verification methods of morphological characteristics, cultural characteristics, chemical component analysis of cell walls, physiological-biochemical characteristics, 16S rDNA complete-sequence analysis, DNA-DNA hybridization, and the like, the similarity between the bacterial strain and the 16S rDNA of the known streptomycete is smaller than 99.0%, the DNA autoploidy is smaller than 50%, and a result indicates that the bacterium species belongs to a novel species of the streptomycete. The bacterium species is suitable for the biological repair of a polluted environment, particularly to an environment polluted by Pb.

Description

technical field [0001] The invention relates to an actinomycete, in particular to a new species of actinomycetes resistant to heavy metal lead, which belongs to the genus Streptomyces and is applied to bioremediation of heavy metal polluted environments. Background technique [0002] Soil is an important part of the earth's ecological environment and one of the important non-renewable resources on which human beings depend. The high concentration of heavy metal pollutants in abandoned land is one of the important environmental pollutants, which aggravates the deterioration of the soil environment. The methods currently used for heavy metal treatment mainly include traditional physical and chemical methods and bioremediation methods. Bioremediation is the use of biological metabolic mechanisms to absorb, precipitate, degrade or redox a certain concentration of toxic and harmful substances. Therefore, bioremediation is more effective for low-concentration pollutants. In addit...
