Abstract
Chimeric replicons of North American Porcine Reproductive and Respiratory Syndrome Virus (PRRSV) containing the 5' sequence of an avirulent strain of PRRSV and a 3' sequence of a virulent strain of PRRSV are provided. Further provided is a method of producing attenuated PRRSVfrom the chimeric replicon. Also provided are compositions containing the replicon or attenuated virus. Vaccines and a method of vaccinating pigs against PRRSV are also provided.

Background technique [0002] Porcine Reproductive and Respiratory Syndrome (PRRS) is the most significant disease economically affecting the swine industry, with the virus causing approximately $5.6 billion in annual losses in the United States. Primary symptoms of the disease are reproductive problems in sows and gilts, including late abortions, stillbirths and mummies, as well as small, frail piglets that are born viremic and often do not survive. In addition, symptoms in young pigs manifest as respiratory disease resulting in fever, lethargy, dyspnea, low appetite, slow growth and occasional death, often in combination with other respiratory pathogens.
