Abstract

The invention relates to a desalting membrane port used in on-line connection with a mass spectrum and the application thereof. The desalting membrane port comprises capillary tubes, a cellulose acetate hollow-fiber membrane and a hermetic container. A sample inlet and a mass transfer exchanging solution outlet, a sample outlet and a mass transfer exchanging solution inlet are arranged on two corresponding sides of the hermetic container. The capillary tubes are sheathed with sample inlet and the sample outlet. The cellulose acetate hollow-fiber membrane passes through and is sheathed among the capillary tubes in the hermetic container through the sample inlet and the sample outlet. An on-line connection system of the desalting membrane port and the mass spectrum comprises a constant flow pump, a mass transfer exchanging solution source, a desalting port, a nano flow mass spectrum spray nozzle and a mass spectrometer. A protein sample is driven by the constant flow pump, goes through the desalting port for desalting, is sprayed by the nano flow mass spectrum spray nozzle and finally is sent to the mass spectrometer for detection. The invention has the advantages of good desalting effect, good repeatability, small dead volume, small loss of protein sample, easy connection with other parts, simple device, convenient operation and low cost.

Description

Technical field [0001] The invention relates to a protein sample pretreatment process, in particular to a membrane interface which can be used in conjunction with mass spectrometry to perform on-line desalination of protein samples. Background technique [0002] Since the 1990s, with the advent of proteomics and the emergence of two ionization methods, electrospray ionization (ESI) and matrix-assisted laser desorption, dissociation ionization (MALDI), mass spectrometry has been used in the process of proteomics research. Become an essential tool. However, because protein samples are very complex and contain many factors that interfere with mass spectrometry analysis, it is very necessary to pretreat protein samples. The pretreatment of protein samples involves a lot of content, including the removal of interference factors such as salt and denaturant, the pre-fractionation of proteins, the enrichment of low-abundance proteins and the removal of high-abundance proteins, and so on....
