Embodiment 1

[0026] ①Raw material processing: Wash 5 kg of fresh Jerusalem artichoke with water, take 2500 grams of fresh Jerusalem artichoke and extract the juice of Jerusalem artichoke with a juicer, use the acid hydrolysis method to determine the total sugar, adjust the total sugar concentration to 10% (w / v), and sterilize at 115 °C After 20 minutes, set aside. Take 2500 grams of fresh Jerusalem artichoke slices, put the Jerusalem artichoke slices into an oven at 70°C, and dry them. After the dried Jerusalem artichoke slices are pulverized by a pulverizer, they are sieved through a 40-mesh sieve to collect fine powder, and the coarse powder continues to be crushed and sieved. The total sugar content of whole Jerusalem artichoke powder, all 40 mesh whole Jerusalem artichoke powders are bottled and sealed for later use.

Embodiment 2

[0031] ①Raw material processing: Wash 10 kg of fresh Jerusalem artichoke with water, slice it, dry the Jerusalem artichoke slices in an oven at 70°C, grind the dried Jerusalem artichoke slices through a grinder, sift through a 40-mesh sieve, collect fine powder, and continue crushing and sieving the coarse powder , combined all 40 mesh whole Jerusalem artichoke powders, and after measuring the total sugar content of the whole Jerusalem artichoke powders by the acid hydrolysis method, the whole Jerusalem artichoke powders were bottled and sealed for later use. Take by weighing about 16 grams of whole Jerusalem artichoke powder and pour it into a 1000ml triangular flask, add 250ml of water, stir evenly, the total sugar content is about 5% (w / v), sterilize at high temperature, and use it as a seed medium for subsequent use.

Abstract

The invention belongs to the biotechnology field, in particular to a method of manufacturing ethanol by utilizing Jerusalem artichoke material. The method comprises the following steps: in material process step, the fresh Jerusalem artichoke is cleaned and juiced, or sliced up, dried and pulverized; in a seed liquid preparation step, kluyveromyces are cultivated in seed culture fluid taking Jerusalem artichoke powders or Jerusalem artichoke juice as a complete composition; in a ferment step, seed liquid is inoculated in culture medium confected by the complete Jerusalem artichoke powders or the complete Jerusalem artichoke juice, the ethanol is produced through anaerobic fermentation, and the ethanol is obtained by distillation at last. Both compositions of a seed culture medium and a ferment culture medium provided by the method are Jerusalem artichoke powders or Jerusalem artichoke juice; any auxiliary composition is not needed; and beforehand saccharification processing of Jerusalem artichoke material in the ferment culture medium is not needed; inulase excreted by the kluyveromyces in the seed liquid directly hydrolyzes and saccharifys multi fructosan in the Jerusalem artichoke material; the ferment is processed at the same time; and concentration of ethanol product is high to 12 percent (v/v). The method has the advantages that the method is simple; the synchronization process of saccharification and ferment is implemented; and the production cost is reduced.

Description

technical field [0001] The invention belongs to the field of biotechnology, and in particular relates to a new ethanol production method. Background technique [0002] Due to the limited and depleted fossil resources, energy shortage has become a major problem facing society today. The development of renewable biomass energy is an effective way to solve the increasingly tense energy supply. Biomass energy is a clean energy product produced from energy plants or organic waste from agriculture and forestry, among which fuel ethanol is the first choice for the development of biomass energy. At present, the annual production scale of fuel ethanol in the world is nearly 30 million tons, while the production capacity of fuel ethanol in my country is only about 1.5 million tons, mainly using corn, wheat and other food crops as raw materials. In view of my country's large population, small land and high pressure on food production, the development space of fuel ethanol using corn,...
