[0019] In accordance with the present invention, there is provided a method of assaying an analyte in a blood sample in which the mean cell volume of the blood sample is reduced to reduce the interfering effect of hematocrit on the accuracy of the assay. According to the method of the present invention, at some point during the diagnostic procedure, the average cell volume of the sample is reduced by treating it with a non-lysing hypertonic saline composition. The term "hypertonic salt composition" as used in this patent means any composition of one or more organic or inorganic salts or mixtures thereof sufficient to create a higher osmotic pressure in a blood sample than isotonic fluid.
