Abstract

The invention discloses a method for establishing an European vaccinium callus regeneration system, which comprises the following steps of: inoculating tender branches of European vaccinium seedlings which grow well under a sterile culture condition as explant materials into a culture medium to obtain explants with calluses, and continuously culturing to generate adventitious buds; the formula of the culture medium comprises WPM, 3% of cane sugar, 1% of agar, 0.5 mg/L IBA and 2.0 mg/L ZT, and the pH is 5.2 +/-0.02; when the adventitious buds grow to 2-3 cm, the adventitious buds are moved to a rooting culture medium for rooting induction. The formula of a rooting culture medium is WPM, 2% of sucrose, 1% of agar, 0.3 mg/L of IAA, 1.0 mg/L of KT and 0.50 g/L of activated carbon, and the pH is 5.2 +/-0.02. According to the method disclosed by the invention, the obtaining rates of the callus and the adventitious buds of the vaccinium myrtillus are respectively 100% and 95% or above. According to the method, only 2-3 months are needed from explants to soil cultivation seedling obtaining, and technical support is provided for clone propagation of vaccinium myrtillus.
