Problems solved by technology

Transfer to these cassettes is complicated and time-consuming, especially when samples were previously housed in the mentioned culture chambers
This is a major obstacle to rapid fixation of cell states in prior art methods and arrangements

[0054] figure 1 A culture chamber 900 for culturing a sample, which does not form part of the invention, is shown in a simplified cross-sectional view and has been described at the outset. Please refer to the instructions above. It should also be noted that the present invention is not intended to be limited to the particular type of culture chamber shown here.

[0055] figure 2 Shown in exploded view is a high pressure freezer box 30, the sample holding means, which may be used with the present invention. Cassette 30 may be used with the above-mentioned high pressure freezers known in the art. For example, reference is made to the literature on the "EM ICE", "EM HPM100" and "EM PACT" systems of Leica Microsystems cited above (e.g. reference to DE 10 2013 003 164 A1 which further describes details of high pressure freezing systems) and to the scientific review literature, For example Kaech & Ziegler (see above). In the assembled state, the cassette 30 can be transferred ...

Abstract

A method is provided comprising one or more steps of a procedure for preparing a microscope sample (1) for a high-pressure freezing process wherein the sample (1) is provided in an arrangement comprising an intermediate plate (33) of a high-pressure freezing cartridge (30) and a culture chamber (100) wherein the intermediate plate (33) is attached by an adhesive to a lower surface of a bottom (120) of the culture chamber (100), the intermediate plate (33) is provided on an enclosure element (37) fitted into an opening (36) of the intermediate plate (33) and is detachable from the culture chamber (100) by effecting a relative movement between the intermediate plate (33) and the culture chamber (100), in which the relative movement between the intermediate plate (33) and the culture chamber (100) is effected when the sample (1) is to be subjected to a high-pressure freezing process, thereby detaching the intermediate plate (33) with the sample (1) from the culture chamber (100). Means for carrying out the method are also part of the invention.

Description

technical field [0001] The invention relates to a method for preparing microscopic samples for high-pressure freezing processes and means for carrying out the corresponding method. Background technique [0002] During light microscopy imaging, living cells should be maintained under favorable environmental conditions. Stage incubators, stage heaters, carbon dioxide chambers, etc. are common tools used for this purpose. [0003] There are several stage incubators and chambers for conditioning the gas atmosphere around samples during light microscopy examinations available. The operating principle of a particular type of culture chamber is disclosed in DE 20 2016 007 488 U1, which offers the option of transferring samples between different locations while still maintaining ambient conditions. [0004] under the figure 1 , the components of this type of culture chamber are shown in longitudinal section. The present invention is not limited to use with any particular type of...
