Problems solved by technology

[0007] There are some methods for detecting PSP, such as biological detection, immunoassay, and chemical analysis, but these methods have defects in accuracy, sensitivity, and poor repeatability, or require large and expensive equipment, professional operators, and complicated and time-sensitive sample pretreatment. However, it cannot be widely used in the monitoring and management of shellfish toxins
In addition, there is currently no toxicity detection technology available for living cells of PSP-producing algae, which seriously restricts the on-site management of shellfish toxins

[0064] The algae were cultured with natural seawater (pH 7.5±0.1, salinity 27‰, purchased from Guangzhou Huangsha Aquatic Products Market), filtered with a 0.45μm microporous membrane, and then sterilized at 121°C and 15psi for 25min , cooled to room temperature. The algal culture solution was prepared with the f / 2 improved formula, placed in an artificial climate box after inoculation, the temperature was 25 °C, and the light intensity was 150 μmolm -2 s -1 , cultured under the condition of light-dark cycle L:D=12:12. Three parallel samples were set up for each algae, and samples were taken at a fixed time every other day. After the algae was fixed, the algal cells were counted under the microscope and the growth curve was drawn.

Abstract

The invention discloses a fluorescence characteristic standard spectrum library for identifying paralytic shellfish poisoning-producing microalgae as well as a construction method and application of the fluorescence characteristic standard spectrum library. The method comprises the following steps of: extracting three-dimensional fluorescence spectrum information of PSP-producing algae and non-PSP-producing algae growing under different environmental conditions, extracting characteristic peaks of experimental algae by using a Db7 wavelet function, performing clustering analysis by using a system clustering method, removing abnormal spectrums, and screening standard spectrums to obtain a Db7 fluorescence characteristic standard spectrum library. When a discriminating function established according to the fluorescence characteristic standard spectrum library is used for discriminating the PSP-producing algae and the non-PSP-producing algae, the discriminating correct rates are 84.6% and95.6% respectively, the correct rates are very high, and the purpose of quickly and accurately identifying the poisoning-producing algae is basically achieved. The fluorescence characteristic standardspectrum library can be used for preparing a red tide algae identification sensor or a portable algae fluorescence identification instrument.

Description

technical field [0001] The invention belongs to the field of identification and prevention of toxic microalgae, and in particular relates to a fluorescent characteristic standard spectrum library for identifying paralytic shellfish toxin-producing microalgae and its construction method and application. Background technique [0002] Red tide refers to the phenomenon that the microorganisms in the water multiply due to the water environment and climate, resulting in the discoloration of the water body. In recent years, the frequency and area of red tides in my country's coastal areas have continued to increase, causing serious economic losses. [0003] According to the toxicity characteristics of red tides, red tides are usually divided into three categories, namely non-toxic red tides, fish toxic red tides and toxic red tides. Among the three types of red tides, the proportion of non-toxic red tides is the highest. This type of red tide does not produce toxins. The toxins ...
