Embodiment 1

[0023] The main purpose of this embodiment is to establish a method for detecting the expression of INSM1 in the sample, the method comprising:

[0024] The kit of this embodiment was composed of INSM1-specific antibody (purchased from Santa Cruz Company, A-8) combined with Maixin S-P immunohistochemistry kit (Fujian Maixin Company). Prostate cancer tissue samples were fixed with 10% paraformaldehyde by mass percentage, dehydrated and transparent with conventional graded alcohol, embedded in paraffin, cut into 4um thick slices, and baked. Perform immunohistochemical staining according to the instructions of the immunohistochemical hypersensitivity kit:

[0025] 1) Put the paraffin sections in a thermostat at 65°C overnight, place them in xylene for 7 minutes, and then soak them for another 7 minutes after replacing the xylenes. Soak in 100% alcohol, 100% alcohol, 90% alcohol, 80% alcohol and 70% alcohol for 5 minutes respectively. Then rinse with tap water twice, each time f...

Embodiment 2

[0035] The main purpose of this embodiment is to evaluate the diagnostic effect of INSM1 expression on the neuroendocrine differentiation of prostate cancer in samples, mainly including:

[0036] 1) Calling 316 subjects whose clinical diagnosis results have been confirmed, including 278 subjects with adenocarcinoma, 27 subjects with adenocarcinoma with neuroendocrine differentiation, and 11 subjects with adenocarcinoma with Paneth cell-like neuroendocrine differentiating subjects;

[0037] 2) Detect the expression levels of INSM1, CHGA and SYP in the samples of 316 subjects respectively;

[0038] 3) According to the diagnostic results of the subjects and the expression levels of different markers in the samples, the sensitivity, specificity and accuracy of different markers were counted;

[0039] 4) Conclusion: According to Table 1, Table 2 and figure 1 , figure 2 The results shown show that the sensitivity, specificity and accuracy of INSM1 in diagnosing adenocarcinoma wi...

Embodiment 3

[0045] The main purpose of this example is to investigate the effect of INSM1 expression on the prognosis of prostate cancer in the sample, mainly including:

[0046] 1) Collect 159 prostate cancer subjects receiving treatment, detect the expression level of INSM1 in the subjects' samples, refer to Example 1 for the specific detection method, and count the clinical progress information of the subjects, including the time of INSM1 detection Check whether the subject has clinical progression and the time when the subject has clinical progression after testing;

[0047] 2) To map the relationship between INSM1 positive expression and INSM1 negative expression and clinical progression;

[0048] 3) Conclusion: According to image 3 The results shown show that the expression level of INSM1 in the sample can predict the clinical progress of prostate cancer and know the prognosis and treatment strategy: detecting the expression level of INSM1 in prostate cancer patients can guide the...

Abstract

The invention belongs to the field of biomedicine, and provides application of insulinoma related protein 1 in preparation of a marker for diagnosis or prognosis evaluation of prostate neuroendocrinesmall cell carcinoma. The invention also provides a kit which comprises a reagent for detecting the insulinoma related protein 1. The marker INSM1 can be used for effectively distinguishing an adenocarcinoma accompanied by neuroendocrine differentiation subject from an adenocarcinoma accompanied by penem cell-like neuroendocrine differentiation subject so that accurate pathological detection of prostate cancer is realized; meanwhile, the marker has a good indication effect in the aspect of adenocarcinoma accompanied by neuroendocrine differentiation diagnosis, and has higher sensitivity and specificity compared with a clinically common diagnosis marker so that the occurrence of clinical misdiagnosis and missed diagnosis is reduced. In addition, the marker also has a good prediction effecton prognosis of prostate cancer, and is beneficial to recurrence monitoring and clinical early intervention of a subject.

Description

Technical field: [0001] The invention belongs to the field of biomedicine, and relates to a diagnostic marker, specifically, the use of insulinoma-associated protein 1 in preparing a marker for the diagnosis or prognosis evaluation of prostate neuroendocrine small cell carcinoma. Background technique: [0002] Prostate cancer is the most common malignant tumor of the male genitourinary system. Its incidence rate ranks first among male cancer patients in the United States, and its mortality rate is second only to lung cancer. According to the latest report released by the American Cancer Society, 191,930 new cases are expected in the United States in 2020, and 33,330 cases will die from the disease. The incidence of prostate cancer in my country is relatively low, but with the aging of the population, the westernization of lifestyle and the popularization of screening methods, the incidence of prostate cancer has shown a significant upward trend. According to the data of the...
